hTNFA-hPgnK5融合蛋白的原核表达及测定 被引量:3

摘要 目的 获得 h TNFα- h Pgn K5融合蛋白基因 ,在原核细胞中表达融合蛋白 ,探索其活性 .方法  PCR法改造h TNFα基因 ,消除其终止密码子 ,构建 h TNFα- h Pgn K5融合蛋白基因 .在大肠杆菌 DH5α中表达该融合蛋白 ,测定其体外抑制肿瘤细胞或血管内皮细胞增殖的活性 .结果  h TNFα基因经 PCR法改造 ,去除了终止密码子 ,并在 3 端引入 Bam HI位点 .h TNFα- h Pgn K5融合蛋白基因构建完成 ,并在原核系统中获得表达 ,免疫印迹反应表明该融合蛋白具有 h Pgn K5免疫活性 .体外实验表明该融合蛋白可抑制肿瘤细胞和血管内皮细胞的增殖 .结论  h Pgn K5蛋白可与 h TNFα一起表达 。 AIM To obtain the gene of hTNFα hPgnK5 fusion protein, express the fusion protein in prokaryotic system and research its characterization.
