摘要
目的 探索大鼠成骨细胞 (OB)在体外不同时间的形态及功能变化。方法 在为期 2 8天的细胞培养中动态观察了新生大鼠头盖骨OB的活细胞形态变化 ,并与其皮肤成纤维细胞 (FB)的形态变化作了比较 ,测定了OB中Ⅰ型原胶原羧基端前肽 (PICP)、骨钙素 (BGP)和碱性磷酸酶(ALP)的表达量变化。结果 OB与FB在早期散在分布时形态相似 ,汇合以后两者逐渐出现差异 ;OB在 2 8天才可用VonKossa法染到明显钙盐沉着 ;PICP、BGP分别于初始植入后第 7、19天有表达高峰 ,ALP在 12~ 16天和≥ 2 8天有两个表达高峰。
Objective To study the temporal changes of morphology and function of cultured osteoblasts from rat calvaria in vitro.Methods The morphology in different culture time of osteoblasts from neonatal rat calvaria and fibroblast from rat skin was compared,and the expression level of procollagen type Ⅰ carboxy terminal propeptide (PICP),Osteocalcin (BGP)and alkaline phosphatase (ALP) were longitudinally investigated.Results When OB and FB were in disperse distribution at early time,their features were very similar to each other,but after confluence,the difference between the two kinds of cells gradually emerged;the calcium nodule could not be detected until day 28 with Von Kossa's staining method;the maximal expression of PICP and BGP occurred on day 7 and 19 respectively,two expression peaks of ALP were observed,one was on day 12~16,and the other was on day 28 or later.Conclusion the morphology and function of rat calvarial OB in vitro have dynamic changes and these cells have the tendency to behave unanimously.
