摘要
目的 :建立适于普遍应用的丙戊酸血浓度测定方法。方法 :本文采用BrMMC将丙戊酸转化为可紫外检测的酯 ,再经RP HPLC测定。结果 :在 15~ 2 0 0 μg·ml-1范围内 ,丙戊酸衍生物和内标的峰高比与浓度呈良好的线性关系 ,r =0 .9991;平均回收率为 10 0 .9%。应用于 3 4例患者的治疗药物监测 ,取得良好的临床效果。结论 :方法准确、灵敏、选择性强、重现性好 ,操作简便快速。本法适于临床研究与应用。
OBJECTIVE To establish a reversed phase HPLC method for routine analysis of valproic acid in human serum.METHODS A reversed phase HPLC method is applied with 4 bromomethyl 7 ethoxycoumain as derivative reagent which transformed valproic acid into an ester detectable with UV detector.RESULTS The ratio of peak heights correlated linearly with concentration of valproic acid ranging from 15 to 200 μg·ml -1 , r = 0.9991 . The mean recovery is 100.9% . The method has been used for therapeutic drug monitoring of 34 patients with epilepsy and satisfied clinical effects is acquired.CONCLUSIONS The method is sensitive, accurate, selective, simple and fast, and suitable for the clinical research and application.
