摘要
目的 :采用薄层色谱衍生化法建立一个快速测定人血清中丙戊酸浓度的方法。方法 :采用正庚酸为内标 ,以α 溴代苯乙酮为衍生化试剂 ,用TLC法分离 ,GF2 5 4硅胶板 ,正己烷∶丙醚 =8∶2为展开剂。结果 :在浓度为 10~ 14 0mg·L-1呈线性关系 ,r =0 .9973 ,回收率为 ( 10 0 .3± 3 .2 ) %。日内和日间的RSD分别为 2 .94 %和 3 .81%。结论 :薄层色谱衍生化法快速测定血清中丙戊酸浓度 ,为临床药物浓度监测提供方法。
OBJECTIVE To establish a method for rapid determination of sodium valproates in serum by TLC after derivatization.METHODS Heptanoic acid was used as an internal standard. The derivatization of sample with α bromo acetophenone is performed after serum extracted with a simple step. The separation was achieved by TLC. The TLC plate was coated with silica gel GF254 using Hexane Isopropyl ether as developing solvent.RESULTS The resultes show that the extraction caliberation curve is linear over the range of 10~140 mg·L -1 ( r = 0.9973 ). The average recovery was ( 100.3 ± 3.2 )% with RSD 2.94 % and 3.81 % for within day and day to day test respectively. CONCLUSIONS This method is good for valproates identfication and clinical monitoring.
