白血病微卫星杂合性缺失的研究 被引量:3

摘要 ①目的 检测白血病微卫星杂合性缺失 (LOH)频率 ,探讨LOH在白血病发病中的作用。②方法利用合成的 11,12号染色体上的 10个微卫星位点引物进行PCR扩增 ,用聚丙烯酰胺凝胶电泳检测法分析 30例不同类型白血病病人的LOH .③结果  30例白血病病人中 ,有 13例发生了 1个以上位点的LOH ,约占 43.3% .其中18例急性白血病病人中有 12例发生LOH ;12例慢性白血病病人中仅有 1例出现LOH ,并于检出后 6 0d发生了急粒变 ,二者LOH频率比较 ,差异有显著性 (P =0 .0 19)。④结论 LOH是急性白血病发病过程中的频发事件 。 Objective To detect the frequency of microsatellite loss of heterozygosity(LOH) and investigate the function of LOH during the progress of pathogenesis in leukemia. Methods Using PCR amplificaton of 10 microsatellite loci at chromosome 11p and 12p in 30 patients with different type leukemia, the production of LOH was analyzed by electrophoresis. Results 13 of 30 patients demonstrated LOH at one or more loci(43.3%). 12 of 18 patients with acute leukemia showed LOH, but only one informative patient(1/12) with chronic leukemia showed LOH (at two microsatellite loci) and this patient occurred acute myeloid leukemia 60 days later. The frequency of LOH in acute leukemia was markedly higher than that in chronic leukemia(P=0.019). Conclusion Microsatellite LOH is a frequent event in acute leukemia and detecting LOH may be an effective screening method for searching candidate tumor suppress gene related to acute leukemia.
