外源性雌激素对大鼠认知障碍的治疗作用 被引量:2

A STUDY OF TREATMENT ON COGNITION DISORDER OF RATS BY USING ESTROGEN SUPPLEMENT THERAPY

摘要 ①目的 了解补充外源性雌激素对大鼠认知障碍的治疗作用。②方法 健康雌性Wistar大鼠 16只 ,随机分为 4组 ,每组 4只。认知障碍模型组 (模型组 ) :应用鹅膏蕈氨酸 (IBA)一次性微量 (1μL)注入大鼠Meyn ert核 ,同时应用D 半乳糖 (4 8mg/kg)皮下注射。认知障碍治疗组 (治疗组 ) :按模型组方法制作认知障碍模型 ,在应用IBA后 7d应用苯甲酸雌二醇 (1mg/kg)和黄体酮 (10mg/kg)治疗 ,每 7d皮下注射 1次 ,共 15 0d .假手术组 :按模型组方法制备模型 ,用人工脑脊液替代IBA ,生理盐水替代D 半乳糖。正常对照组 :不应用任何药物。用MG 2迷宫观察各组大鼠学习记忆成绩 ,苏木精 伊红染色观察大鼠脑组织形态变化。③结果 应用IBA和D 半乳糖 15 0d后 ,模型组大鼠学习记忆能力较其他组明显低下 (F =8.392 ,P <0 .0 1) ,其电击次数 (2 5 .5 0± 12 .39)明显高于治疗组 (3.80± 3.83)、假手术组 (5 .5 0± 7.5 5 )和对照组 (2 .2 5± 4.5 0 ) (q =5 .2 6 1~ 6 .116 ,P均 <0 .0 5 ) ;而治疗组与假手术组和对照组比较差异无显著性 (q =0 .42 9,0 .471,P均 >0 .0 5 )。光镜下模型组海马皮质单个视野中锥体细胞计数 (3.2 5± 2 .6 0 )明显少于治疗组 (15 .0 0± 6 .35 )和对照组 (17.85± 11.19) (t=3.5 13,2 .5 44 。 Objective To observe the effectiveness of the treatment on cognition disorder of rats model by using the estrogen supplement therapy. Methods A total of 16 healthy male wistar rats were randomly divided into 4 groups. The cognition disorder model group: the model was made by a single injection of ibotenic acid (IBA,1μL) into the Meynert nucler using microdrive injector. D galactose(48mg/kg) was used through subcutaneous injection. Once a day, lasted for 150 days. The cognition disorder treatment group: the same method was used to make the disorder as the model group. But the rats were treated with the estrogen supplement therapy(benzoate estradiol 1mg/kg). The artificial group: the rats were injected artificial CSF instead of IBA and NS instead of D galactose. The normal group: without any drugs. The MG 2 maze was used to record the ability of learning and memory. The pathology of brain was observed by HE staining. Results IBA and D galactose could damage the ability of learning and memory. After 150 days of the injection, the times of learning and memory of the model group(25.50±12.39) were more higher than the treatment group(3.80±3.83), the artificial group(5.50±7.55) and the control group(2.25±4.50) (q=6.017,5.261,6.116,P<0.05). The average amounts of neuro cells in one visual field were more decreased in the model group(3.25±2.60) than the treatment group(15.00±6.35) and the control group(17.85±11.19), (t=3.513,2.544,P<0.05). But the treatment group had no difference with the control and artifical groups (t=0.450,0.120;P>0.05). More Alzheimer Ⅱ cells at the hippocampus were observed in the model group. But there was no more difference between the model and the treatment groups. Conclusion The estrogen supplement therapy had more efficiency in the treatment of the disease.
