RP-HPLC法测定清热解毒汤剂中葛根素的含量

Determination of Puerarin in Heat clearing Toxin resolving Decoction by RP HPLC

目的 :建立用反相高效液相色谱法测定清热解毒汤剂中葛根素含量的方法。方法 :采用HypersilC18柱 ;流动相为乙腈 2 %冰醋酸溶液 (8.5 :91.5 ) ,流速为 0 .8ml·min 1,检测波长为 2 5 0nm。结果 :葛根素对照品线性范围 0 .42 4～4.2 40 μg,r=0 .9998,平均回收率 10 1.3 % ,RSD =1.4%。结论 :本方法操作简便、准确、重复性好 ,可作为该制剂的定量分析方法。

Objective: To establish a RP HPLC for determination of puerarin in Heat clearing Toxin resolving Decoction. Methods: A Hypersil C 18 column with acetonitrile 2% glacial acetic acid (8.5:91.5) as a mobile phase was used. The flow rate was 0.8ml·min 1 and the detective wavelength was 250nm. Results: The calibration curve was linear at a range of 0.424 4.240μg for the puerarin (r=0.9998). The average recovery was 101.3% and the relative standard deviation (RSD) was 1.4%. Conclusion: The methld is simple, accurate with a good reproducibility and can be used as a quantitative analysis method for this preparation.
