枸橼酸铋钾对预防乙醇所致大鼠急性胃粘膜损伤的实验研究 被引量:5

摘要 目的 :研究枸橼酸铋钾对乙醇所致大鼠急性胃粘膜损伤的保护作用及其机制。方法 :将 46只健康雄性Wistar大鼠随机分为 4组 :空白对照组 (Ⅰ ) ;单纯损伤组 (Ⅱ ) ;硫糖铝预防保护组 (Ⅲ ) ;枸橼酸铋钾 (商品名为丽珠得乐 )预防保护组 (Ⅳ )。分别用硫糖铝和丽珠得乐胶囊提前给预防保护组大鼠灌胃 ,其后用乙醇灌胃致急性胃粘膜损伤 ,然后分别测定各组大鼠的胃粘膜溃疡指数、胃粘膜血流、胃粘膜氨基己糖含量及前列腺素含量等指标 ,并在显微镜下观察组织学变化 ,以评价胃粘膜损伤程度及药物预防保护效果。结果 :实验测得Ⅰ ,Ⅱ ,Ⅲ ,Ⅳ各组的胃粘膜损伤指数依次为 0 .46± 0 .6 9,5 7.5 5± 18.81,9.17± 3.5 4,3.6 7± 2 .6 4;胃粘膜血流依次为 46 .0 0± 6 .5 1,39.46±7.16 ,43.83± 3.86 ,5 0 .83± 6 .5 8(胃窦 ) ;39.6 4± 5 .6 4,38.36± 9.0 3,46 .83± 7.36 ,5 3.83± 7.6 5 (胃体大弯 ) ;5 1.2 7±6 .34 ,44 .91± 5 .39,48.83± 6 .5 8,5 3.83± 6 .95 (胃体小弯 ) ;胃粘膜氨基己糖含量 (μg·mg-1蛋白 )依次为 118.2 3±15 .82 ,6 7.5 2± 12 .0 0 ,82 .6 3± 13.95 ,136 .2 2± 2 1.6 0 ;胃粘膜前列腺素含量 (pg·mg-1蛋白 )依次为 42 .37± 5 .38,11.94± 1.96 ,2 1.95± 3.84,5 8.82± 9.0 Objective:To study the protective effect of bismuth potassium citrate(BPC) on gastric mucosa in rats'model.Methods:46 healthy male Wistar rats were randomly divided into four groups :I(control group),Ⅱ(injury group),Ⅲ(sucralfate protective group),and Ⅳ(BPC protective group).The two protective groups were treated with sucralfate and BPC respectively before to induce gastric muscle injury.Then the ulcer index and blood flow of gastric mucosa,and the concentration of hexamine and prostaglandin were measured and the pathological changes were observed under microscope.Results:The data obtained were as follow:ulcer index:0.46±0.69,57.55±18.81,9.17± 3.54, 3.67±2.64 respectively;blood flow:46.00±6.51,39.46±7.16,43.83±3.86,50.83±6.58(pyloric antrium);39.64±5.64,38.36±9.03,46.83±7.36,53.83±7.65(greater curvature); 51.27± 6.34,44.91±5.39,48.83±6.58,53.83±6.95(lesser curvature) respectively;hexamine concentration(μg·mg -1 protein):118.23±15.82,67.52±12.00,82.63±13.95,136.22±21.60 respectively;and prostaglandin concentration (pg·mg -1 protein):42.37±5.38,11.94±1.96,21.95±3.84,58.82±9.08 respectively.In comparison with injury group,in BPC protection group the ulcer index of gastric mucosa were significantly decreased( P <0.001),the blood flow of gastric mucosa were significantly increased( P <0.05),hexamine concentration were significantly increased( P <0.001) and prostaglandin concentration were significantly increased( P <0.001).The pathological observation indicated that a negligible degree of inflammation and injury were observed in BPC protection group. Conclusion: The data obtained revealed that BPC has a significant protection effect on rats with acute gastric mucosa injury induced by ethanol and the increase in blood flow,and hexamine and prostaglandin concentration may be contributed to the protective effects.
