摘要
目的 :探讨内源性EGF促进深二度烫伤创面愈合的作用机制。方法 :采用免疫组织化学染色、核酸分子原位杂交和图像分析技术 ,动态检测摘除和不摘除颌下腺小鼠深二度烫伤创面愈合过程中 ,创面组织和颌下腺EGF/EGFR及其mRNA的表达部位及其规律与强度。结果 :创面组织中EGF/EGFR及其mRNA均表达于毛囊上皮细胞、成纤维细胞和血管内皮细胞 ,其表达强度呈现不同的规律性。颌下腺组织中EGF主要表达于导管系统 ,伤后 3天内的表达强度明显低于伤前水平 (P <0 .0 1 ) ,伤后 5天基本恢复至伤前水平 (P >0 .0 5) ;EGFmRNA的表达部位与EGF基本一致 ,但伤前伤后表达强度无明显变化。结论 :内源性EGF借助于自分泌、旁分泌和内分泌机制促进了深二度烫伤创面愈合。
Objective:To study the mechanism of action to accelerate second deep degree scald wound healing by endogenous EGF.Methods:During second deep degree scald wound healing in mice with or without submaxillary gland,immunohistochemistry,hybridization and image analysis were employed to dynamically detect the sites,regularities and intensities of EGF,EGFR and their mRNA expressions in wounds and submaxillary gland.Results:The expressed EGF,EGFR and their mRNA in wounds appeared in the same sites,epithelial cells of hair follicle,fibroblasts and endothelial cells,and their expressed intensities showed different regularities.In submaxillary gland,the expressed EGF mainly appeared in duct system,its expressed intensity showed a marked decrease compared with the control in three days post-injury( P <0.01).On the 5 th day,the expressed intensity reached the same level as that of control( P >0.05).The expressed EGFmRNA appeared in the same sites with expressed EGF,but there was no change in expressed intensities between pre-injury and post-injury.Conclusion:Endogenous EGF might accelerate second deep degree scald wound healing by the autocrine,paracrine and endocrine mechanism.
