摘要
探讨阿魏酸钠对TNF α致血管内皮细胞 (VEC)中Bcl 2和Bax表达改变的影响及其细胞保护作用。以人脐静脉内皮细胞 (HUVEC)为实验模型 ,采用细胞培养、电镜、免疫组化等技术观察TNF α对VEC的形态以及内皮细胞中Bcl 2和Bax表达的影响 ,并研究阿魏酸钠的保护机理。结果 ,TNF α能明显损伤VEC的超微结构 ,可见较多凋亡细胞 ,细胞中Bcl 2表达明显减少 ,Bax表达明显增多。将阿魏酸钠与TNF α共同作用于VEC ,可见内皮细胞的超微结构与正常细胞形态相似 ,Bcl 2表达增多 ,Bax表达减少。阿魏酸钠可减少TNF α导致内皮细胞的损伤和凋亡 ,其机理可能是通过减少Bax表达 ,提高Bcl
Objective:The aim of this article is to study the effects of sodium ferulate on the expressional changes of Bcl 2 and Bax in the vascular endothelial cells (VEC) induced by TNF α and the protective effects of TNF α on the cells.Methods:In this study,human umbilical vein endothelial cells (HUVEC) were used as the target cell,and the methods of electron microscope and immunocytochemistry were used to observe the changes of microstructures and the expressions of Bcl 2 and Bax in VEC induced by TNF α.Results:In the presence of TNF α,the microstructure of VEC were significantly damaged,and the expressions of Bcl 2 and Bax in VEC were significantly decreased and increased respectively.We also have found many apoptosis cells in the study.But when we added the sodium ferulate to the culture medium of VEC in the presence of TNF α,We found that the microstuctures of the cells were as the same as the normal cells,and the expressions of Bcl 2 were increased,the expressions of Bax were decreased.Conclusion:Sodium ferulate can reduce the damnification and apoptosis of endothelial cells induced by TNF α.The effects of sodium ferulate on the expressions of Bcl 2 and Bax may be the underlying mechanism.
