摘要
用感染了柯萨奇A16(CA16)病毒的1~2日龄乳鼠的肌肉浸出液制备该病毒抗原,建立了抗体捕捉ELISA法(MacELISA),并检测59份手足口病(HFMD)患者血清CA16-IgM抗体.其中16例为急性期和恢复期双份血清,经MacELISA法检测,13例CA16-IgM阳性(81.25%),间接ELISA法检出CA16-IgG抗体4倍升高者12例(75%)。此12例又经MacELISA法检测,11例CA16-IgM阳性,检出率达91.7%。病后2~4天IgM检出率约为80%(28/36),5~25天达100%(10/10),26~31天降为33.3%(2/6),32天后测不出(0/8)。本方法特异性高。重复性良好,可作为CA16病毒感染的快速、特异的早期诊断方法。
Using the Coxsackie Al6 virus antigens extracted from the muscles of infected suckling mice, we developed an IgM capture ELISA test for the detection of Coxsackie Al6 virus infection. 27 cases with single sera from hand-foot-mouth disease ( HFMD ) and 16 cases with paired sera from HFMD were tested. Of the 16 cases with paried sera, 13 were CAl6-IgM positive by MacEI.ISA and 12 had IgG 4-fold rise by indirect ELISA. Among the 12 cases with IgG 4-fold rise, the IgM antibody detection rate was 91.67%(11/12) . The specific IgM antibody was detected on second day after onset of the illness. About 80% of the patients were IgM-posi-tive on days 2-4.All the patients were IgM-positive on day 5-25,and then the IgM antibody decreased. 32 day after the illness, no IgM-positive cases were detected. The MacELISA for the detection of CAl6-IgM is a rapid, sensitive and simple method for speific clinical diagnosis.
