摘要 目的 :研究重组人肝细胞生长因子 (rh HGF)对常染色体显性遗传型多囊肾病 (ADPKD)囊肿衬里上皮细胞增殖及细胞外基质合成的影响。方法 :用 3H- Td R掺入法测定不同浓度 (0 .5、1、2 .5、5 ng/m l) rh HGF作用 48h以及最适浓度的 rh HGF分别作用 2 4、48、72 h ADPKD囊肿衬里上皮细胞系细胞的增殖情况 ;并分别用 3H -脯氨酸掺入法和放免法测定最适浓度rh HGF作用最佳时间后 ADPKD囊肿衬里上皮细胞系细胞胶原及层粘连蛋白的合成情况。 结果 :rh HGF促进了 ADPKD囊肿衬里上皮细胞增殖及胶原和层粘连蛋白的合成 ,以 1ng/m l持续作用 48h最为显著。结论 :rh HGF对体外培养的 ADPKD囊肿衬里上皮细胞增殖及细胞外基质合成有明显的促进作用。 Objective: To study the influence of recombinant human hepatocyte growth factor (rhHGF) on proliferation and extracellular matrix synthesis of autosomal dominant polycystic kidney disease (ADPKD) cyst lining epithelial cells in vitro . Methods: The effects of different concentration rhHGF (0.5,1,2.5,5 ng/ml)in 48 h and the optimal concentration rhHGF of different time (24,48,72 h ) on proliferation of ADPKD cyst lining epithelial cell lines were observed by the incorporation of 3H TdR, and synthesis of collagen and laminin were respectively observed by the incorporation of 3H proline and radioimmunoassay. Results: rhHGF stimulated the proliferation of ADPKD cyst lining epithelial cells and synthesis of collagen and laminin,the optimal concentration and time of rhHGF were 1 ng/ml and 48 h. Conclusion: rhHGF can significantly stimulate ADPKD cyst lining epithelial cells proliferation and extracellular matrix synthesis in vitro . [
