摘要
目的 观察金属蛋白酶抑制剂BB 94对涎腺腺样囊性癌 (adenoidcysticcarcinoma ,ACC)细胞侵袭、转移能力的抑制作用。方法 癌细胞体外侵袭能力用重组基底膜侵袭实验测定 ,聚丙烯酰胺凝胶电泳 (polyacrylamidegelelectrophoresis,PAGE)底物酶谱法检测Ⅳ型胶原酶活性 ,用腺样囊性癌ACC M细胞系裸鼠肺转移模型观察体内抑制转移作用。结果 在 10 μmol/L和 10 0 μmol/L时 ,BB 94对ACC M细胞侵袭重组基底膜的抑制率分别为 32 9%和 5 3 2 % ;对ACC M细胞培养上清中的Ⅳ型胶原酶活性有降低作用。在裸鼠肿瘤转移实验中 ,对照组和用药组的肺转移率分别是 90 0 %和2 0 0 % ,P <0 0 1;含转移灶肺重量分别为 (0 2 0 90± 0 0 6 6 7)g和 (0 15 32± 0 0 378)g ,P <0 0 5。结论 金属蛋白酶抑制剂BB 94能明显抑制ACC M细胞侵袭重组基底膜和ACC的肺转移。
Objective To study the inhibitory effect of matrix metalloproteinase inhibitor BB-94 on t he invasion and metastasis of salivary adenoid cystic ca rcinoma in vitro and in vivo. Methods Reconstituted basement m embrane invasion assay was used to evaluate invasive ability of cancer cells. Ty pe Ⅳ collagenase was assessed by PAGE substrate zymography. In addition, the an timetastasis effec t of the compound was studied in in vivo experiments using nude mice model o f pu lmonary metastasis with ACC-M line of human salivary adenoid cystic carcinoma. Results BB-94, at the concentrations of 10μmol/L and 100 μm ol/L, suppressed the reconstituted basement invasion of ACC-M human salivary ad enoid cystic carcinoma by 329%, 532% and reduced Type Ⅳ collagenase activit ies in the serum-free supernatant of the same cells. The pulmonary metastasis r ate of the control and treated groups was 900% and 200%, respectively (P <001). The lung weight of the control and treated groups was (0209 0±0 066 7) g and (0153 2±0037 8) g , respectively (P<005). Conclusions These result s suggest that the matrix meta lloproteinase inhibitor BB-94 has a strong inhibitory effect on invasion and me tastasis of salivary adenoid cystic carcinoma.
