摘要
目的 探讨癫痫过程中神经细胞Na+ 、K+ 通透性的变化。方法 应用Na+ 、K+ 选择性微电极检测了马桑内酯 (5 μl,5× 10 -4 mol/L)灌流的大鼠海马脑片神经细胞外Na+ 、K+ 活度的改变。结果 马桑内酯灌流海马脑片 30s后可观察到海马神经细胞外Na+ 活度明显下降 ,与对照组比较下降幅度为 10mmol/L(P <0 .0 1) ,在实验观察的时间内 (2 0min)未见Na+ 活度的恢复 ;海马神经细胞外K+ 活度在灌流后 1min、2min、3min、5min和 10min分别高于对照组 3.3mmol/L、4.8mmol/L、4.9mmol/L、6 .5mmol/L和 8.8mmol/L ,在以后 10min内K+ 活度亦未见恢复。结论 马桑内酯作用于离体海马神经细胞时存在Na+ 内流、K+
Objective To study the changes of ionic activity of extracellular sodium and potassium (o?o) of hippocampal slice perfused by coriaria lactone (CL).Methods The sodium ion (Na +) and potassium ion (K +) selective microeletrodes were used to measure alteration of the o and the o of perfused hippocampal slice with CL. Results 30s after perfusing of CL into hippocampal slice, the o decreased 10 mmol/L compared with control values ( P <0.01) and did not return to the control level during the experiment (20min) .1min ?2min ?3min ?5min and 10min after perfusing of CL into hippocampal slice,The o increased 3.3mmol/L ?4.8mmol/L ?4.9mmol/L? 6.5mmol/L and 8.8 mmol/L respectively compared with control values ( P <0.01), 10 min after, the o always kept the level. Conclusion: The influx of Na + and the flux of K + occurred during perfusing hippocampal slice with CL .
