摘要 目的:了解14株(亚株)人鼻咽癌细胞的放射生物学特性。材料与方法:采用集落存活分析法分析中国建成的14株(亚株)人鼻咽癌细胞的特性;除了CNE1之外,所有的细胞均为低分化鳞癌细胞,实验求得的放射剂量存活曲线用线性二次模型进行拟合,用Fertil 和Malais 介绍的方法和原理计算存活曲线的初斜率(α),平均抑制剂量(mean inactivation dose, MID)及2 Gy处的存活分数(SF2)。结果:α:0.001~0.81, MID:1.11~2.46, SF2:0.17~0.66, 14株(亚株)鼻咽癌细胞的放射敏感性变化较大,来源同一个母株(SUNE1)的8个亚株之间的放射敏感性存在着差别。结论:鼻咽癌细胞在个体内和个体间变化较大;因此,在临床制定鼻咽癌的治疗计划时,要考虑鼻咽癌的个体放射生物学特性。 Objective:This study was designed to determine the radiobiological characteristics of a panel of 14 human nasopharyngeal carcinoma cell lines (sublines). Methods and Materials:Fourteen human nasopharyngeal carcinoma cell lines (sublines) from Chinese patients were characterized using the clonogenic survival assay. All cell lines, except for cell line CNE1, were poorly differentiated squamous cells. A radiation dose-survival curve was obtained for each experiment. All survival data were analyzed using linear quadratic model and then values of the initial slope of the survival curves (α), the mean inactivation dose (mean mactivation dose, MID), and the surviving fraction at 2 gray (SF2) were calculated using the methods and rules described by Fertil and Malaisl. Results:The following ranges of values were obtained:α, 0.001-0.81; MID, 1.11-2.46; SF2, 0.17-0.66, respectively. The radiosensitivities of 14 human nasopharyngeal carcinoma cell lines tested varied widely. Furthermore, radiosensitivities differed among eight sub-lines derived from one parent cell line (SUNE1). Conclusion:The radiosensitivities of nasopharyngeal carcinoma cell lines may vary significantly within and among individuals. Thus, in planning clinical treatment for nasopharyngeal carcimona, individual radiobiological characteristics may be useful for predicting treatment.
