摘要
目的 用紫外分光光度法测定补充不同牛磺酸水平的大鼠和雏鸡脑组织中牛磺酸含量。方法 1将清洁级 SD雄性断乳大鼠随机分为四组 ,每组 8只 ,A对照组 ,B补充 0 .6 %牛磺酸 ,C补充 1.35 %牛磺酸 ,D补充 3.0 %牛磺酸。另将 1、3、8周龄海兰褐雏鸡随机分为四组 ,每组 5只 ,各组补充牛磺酸的量与大鼠相同。喂饲4周后 ,处死动物 ,取出脑组织存于 - 70℃待测。2用离子交换色谱法分离提取脑组织中的牛磺酸 ,用紫外分光光度法测定牛磺酸含量。结果 补充牛磺酸的大鼠及雏鸡脑牛磺酸含量显著高于对照组 (P<0 .0 5 ) ,测得方法回收率范围为 92 .5 %~ 10 5 .5 % ,最佳吸收波长为 35 5 nm。结论 该方法快速、简便而且重现性好 ,不受大型仪器限制 。
Objective To determ ine the content of taurine in rats and chickens′brain,which had been fed by different level taurine.Methods 1Male weanling SD strain rats were divided into four groups,eight rats per group.Group A was given control diet,groups B,C and D were given on of the control diets incorporated with taurine at the levels of0 .6 % ,1.35 % or3% respectively.At the same time,1,3,8w old Hailang chickens were divided into four groups,five chickens per group.Taurine content of each group was same as rats′.The experi- ment lasted four weeks.At the end of experiment,brain of animals were stored at - 70℃ for use.2 Taurine was isolated from brain tissues with ion exchange chromatography,and then its content assayed by ultraviolet spec- trophotom etry.Results All levels of taurine in brain of rats and chickens were significantly higher than those in control group(P<0 .0 5 ) .The m ethod had a recovery rate92 .5 %~ 10 5 .5 % ,with a best absorption wavelength 35 5 nm.Conclusion The method is rapid,sim ple,with a good reproducibility,without need of any large equip- menti so it can be used in general laboratory.
