摘要 目的 :探讨食管癌组织中端粒酶逆转录酶 (hTRT)的表达和端粒酶活性的关系。评价端粒酶hTRT表达检测在食管癌诊断中的价值。方法 :用免疫组化和TRAP法分别观察 5 8例食管癌和 2 8例正常食管组织 ,比较分析它们的相关性。结果 :5 8例食管癌hTRT阳性率 83% (48/ 5 8) ,端粒酶活性阳性率 86 % (5 0 / 5 8) ,2 8例正常食管组织均为阴性。中低分化的食管癌端粒酶hTRT及端粒酶活性阳性率高于高分化食管癌。结论 :抗hTRT抗体在食管癌中具有癌细胞的特征性 ,并与端粒酶活性呈正相关。随组织学分级的增高 ,端粒酶hTRT及端粒酶活性阳性率增高。提示其检测有望成为预测肿瘤发生、肿瘤诊断的良好指标。用免疫组化法比TRAP法检测食管癌中端粒酶的活性将更为简便易行。 Purpose:To investigate the expression of human telomerase reverse transcriptase (hTRT) in esohageal carcinoma and the relationship with the elomerase activity and to evaluate the role of detection of hTRT in esophageal carcinoma diagnosis.Methods:58 esophageal carcinoma and 28 normal esophageal tissues were analyzed by immuno-histochemistry for the hTRT and the telomerase activity was measured by TRAP assay.Results:The TRAP assay showed 86%(50/58) of the carcinoma cases to be positive, sample which presented a telomerase activity; and the expression rate of hTRT is 83%(48/58),and 28 normal are negative. The frequency of hTRT and telomerase expression differed significantly between well/moderately differentiated and poorly differentiated tumors.Conclusions:hTRT,as an index of esophageal cacinoma,shows it to be valuable in esophageal carcinoma diagnosis, and the immunohistochemistry of hTRT is easier than TRAP assay.
