端粒酶hTRT基因在睾丸肿瘤组织中的表达及其意义 被引量:9

摘要 目的 研究端粒酶hTRT基因在睾丸肿瘤组织的表达及其意义。方法 应用核酸原位杂交技术对 5 1例男性睾丸肿瘤组织和 10例正常睾丸组织中端粒酶hTRT基因的表达进行检测和定位 ,并应用HPIAS 10 0 0高清晰度图像处理系统对hTRT阳性信号进行图像分析。结果 端粒酶hTRT基因在睾丸肿瘤组织中有极高的表达 ,其阳性率为 92 .16 % ( 4 7/5 1) ,端粒酶hTRT基因表达强度与肿瘤细胞的分化程度显著相关 ,睾丸良性肿瘤与睾丸恶性肿瘤间差异有显著性(P <0 .0 5 ) ,在睾丸肿瘤组织和癌旁组织及正常睾丸组织中的表达强度差异有非常显著性(P <0 .0 1) ,并且其强阳性表达水平与肿瘤细胞的分布定位一致。结论 端粒酶hTRT有可能成为睾丸肿瘤诊断的新标志物及治疗的新靶点。 Objective To study the expression of telomerase gene hTRT in human testicular tumors and its significance.Methods Using in situ hybridization(ISH) techniques, the expression of telomerase gene hTRT mRNA in 51 human testicular tumors and 10 normal testicular tissues was detected. The hTRT positive signaling was analyzed by using HPIAS 1000 high resolution imaging processing system.Results The expression of hTRT was significantly higher in testicular tumors than that in paracancer and normal testicular tissues(P<0.01),and the positive percentage was 92.16%(45 of 51).There was a positive correlation between the expression of hTRT and the location of tumor cells.Moreover,telomerase hTRT expression was also correlated with tumor cell differentiation,and the magnitude of the expression was statistically defferent from benign and malignant testide neoplasm(P<0.05).Conclusion Detection of hTRT was valuable to clinical oncology diagnosis and hTRT might be a new pathway for the gene therapy of testicular tumor.
