摘要
目的 通过检测肾癌相关新基因GYLZ RCC1 8(基因号 :BE82 51 3 3 )在肾癌中的表达特点及其对肾癌细胞生长、增殖的影响 ,探讨GYLZ RCC1 8基因的功能及其在肾癌发生发展中的作用。方法 应用逆转录 聚合酶链式反应 (RT PCR)的方法检测肾癌相关基因GYLZ RCC1 8在3 1例肾癌标本中的表达特点 ;观察GYLZ RCC1 8反义寡核苷酸对肾癌细胞生长、增殖活性、形态学的影响。结果 GYLZ RCC1 8基因在肾癌组织特异表达 ,正常肾组织较肾癌组织低表达或不表达 ,两者表达量差异为 1~ 9倍。GYLZ RCC1 8基因的表达在高分级、高分期肾癌明显高于低分级、低分期肾癌 ;导入GYLZ RCC1 8反义寡核苷酸后 ,可明显抑制癌细胞的生长和增殖活性 ,并减少GRC 1细胞的核分裂。结论 GYLZ RCC1 8是肾癌组织相对较特异表达的新基因 ,它的高表达与肾癌的发生发展以及生长、增殖密切相关 ,它的成功克隆为阐明肾癌发生发展的分子生物学机制开辟了新的途径 ,为今后肾癌的特异诊断 ,基因治疗提供了新的理论依据。
Objective To study the expression of novel gene GYLZ RCC18 in renal cell carcinoma (RCC) and normal kidney tissue and the role of GYLZ RCC18 in generation and development of RCC cell line GRC 1.Methods By using RT PCR technique the expression of GYLZ RCC18 in differential grades and stages was detected.Also 20?bp GYLZ RCC18 antisense oligonuclotide packed with liposome was transfacted into GRC 1 cell and the changess in growth speed, proliferation activity, and morphology of GRC 1 observed.Results GYLZ RCC18 was expressed specially in RCC in higher level than in normal kidney tissue.GYLZ 18 overexpressed in high grades and high stages samples,9 times more than lower grades and stages ones. After transfection of GYLZ RCC18 antisense oligonuclotide the mortality of GRC 1 was increased evidently,and the proliferating activity and growth speed were inhibited remarkably.Conclusions GYLZ RCC18 is a novel gene which is specially relative with RCC.The overexpression of GRLZ RCC18 has a close correlation with the generation and development of RCC.Its successful cloning would provide a new path to research the molecular mechanism of the generation and development of the RCC and the direction of the special diagnosis and therapy for RCC.
