摘要
从10例人腮腺瘤活检组织的冰冻切片及组织培养物中检测到了EB(Epstein-Barr)病毒相关抗原;从活检组织培养上清中分离的EB病毒对淋巴母细胞有感染活性。用电镜观察,在腮腺上皮细胞中可见到EBV样颗粒。提示,腮腺是EB病毒持续感染的组织,是口腔唾液中EB病毒的来源之一。用组织培养上清分离的EB病毒感染7例人胚鼻咽上皮细胞后,能表达EB病毒相关抗原(EBNA和VCA)。讨论了来自腮腺组织的EBV在鼻咽癌发病机制中的可能作用。
Human parotid gland tissues were cultivated according to the method developed in our laboratory and the morphology of cultivated cells under phase-contrast microscope or electron microscope as well as the expression of EBV-specific antigen in the cultivated cells were examined. Theseex-periments show that the cells cultivated are epithelial cells and express EBNA and/or VCA antigen specific for EBV. Some EBV-like partictes were found in the cytoplasm of degenerated cells. Taken together, human parotid gland cells may be considered as permissive cells for EBV. The concentrated filtrate of culture medium of human parotid gland cells was examined under EM for EBV by negative staining and inoculated into Raji,Ramos and cultivated human embryonic nasopharyngeal epithelium in order to assess its infectivity. The results showed that EBV-like particles were found in the concentrated filtrate and EBNA, VCA tests were positive in human embryonic nasopharyngeal epithelium after the cells were exposed to the filtrate. Therefore,the human embryonic nasopharyngeal epithelium may be infected directly by EBV derived from the parotid gland. The data thus obtained indicate that the EBV derived from the parotid gland may play an important role in the pathogenesis of nasopharyngeal carcinoma.
