摘要
为阐明酵母PHO81基因与酸性磷酸脂酶(APase)家族中其他基因的相互关系,用已构建的PHO2、pHO85、PHO5、PHO11及PHO81基因与半乳糖音酶基因(LacZ)的重组质粒对野生型酵母菌和PHO2、PHO85、PHO81等调控基因突变株分别转化,以β-半乳糖苷酶的活力表示各基因的表达水平,分析这些调控基因对其它基因表达的影响。结果表明,PHO81基因对结构基因的表达起着正调控作用。调控基因PHO2、PHO85的表达不受PHO81基因的影响,而PHO81基因的表达却受前者的调控。在低磷条件下,PHO2基因的突变使PHO81表达水平下降7倍,PHO85的突变则使PHO81呈构成型高效表达。
PHO81 gene has an important role in the regulation and expression of the yeast acid phosphatase.To study the relationship between PHO81 gene and other genes of APase fami-ly,the fusion genes.such as PHO2-Lac Z、PHO85-Lac Z、PHO5-Lac Z、PHO11-Lac Z,Weretransferred into the wildtype and PHO81 gene mutation yeast strains respectively.ThePHO81-LacZ was also transfered into wild type and PHO2 or PHO85 mutation strains.Theresult showed that PHO81 gene has positive regulatory role on the structure genes but thegene irself regulated by PHO2 and PHO85 genes.From above,a new APase regulatory mod-el is proposed.
