用2-氯-4-硝基苯-α-半乳糖麦芽糖苷作底物测定总淀粉酶和胰腺淀粉酶 被引量:4

摘要 目的 建立一个稳定的单一试剂直接测定总淀粉酶和胰腺α 淀粉酶方法。 方法 用 2 氯 4 硝基苯 α 半乳糖麦芽糖苷 (Gal G2 α CNP)作底物 ,同时用麦芽唾液淀粉酶抑制物直接测定总淀粉酶和胰腺淀粉酶。淀粉酶水解Gal G2 α CNP为半乳糖麦芽糖苷和CNP ,在 4 0 5nm波长测定。 结果 本法不需要其它工具酶 ,延迟相较短 ( <60s) ,线性范围可达 1 0 0 0U/L ,并且试剂的稳定性很好。精密度分析批内CV<2 .2 5% ,总CV <3 .0 5。本法 (Y)与IFCC推荐方法 (X)比较 :r=0 .9997,Y =1 .3 74 5X +0 .0 4。 结论 本法测定体液中总淀粉酶和胰腺淀粉酶活性非常简便和灵敏 ,适用于常规分析。 Objective To build a method for direct measuring total amylase and pancreatic amylase. Methods We have used 2 Chloro 4 nitrophenyl 4 0 D galactopyranosylmaltoside (Gal G 2 α CNP) as the substrate and wheat germ ptyalin inhibitor for determination of T AMY and P AMY. Amylase converted Gal G 2 α CNP into β D galactopyranosylmaltose and 2 chloro 4 nitrophenol, which was measured at 405nm. Results The method is no need for auxiliary enzymes, the lag phase of this test was < 60 s,the linear range up to 1000 U/L and the reagent was stable. The precision were <2.25% for within run CV, and <3.05% for total CV. Correlation between results obtained by this method (Y) and IFCC (X) was Y=1.3745X + 0.04,r=0.9997. Conclusion This novel assay for measuring stoichiometrical activities of T AMY and P AMY is an easy and direct method, and it may be suitable for routine procedures.
