急性冠脉综合征单核细胞过氧化物酶体增生激活型受体γ基因表达及其与细胞黏附分子相关性研究 被引量:10

A study on peroxisome proliferator activated receptors γ expression in peripheral blood monocyte and correlation with adhesion molecules in acute coronary syndrome

摘要 目的 检测过氧化物酶体增生激活型受体γ(PPARγ)在单核细胞的表达 ,观察急性冠脉综合征时PPARγ的表达状态及其与细胞黏附分子的相关性。方法 从 43例急性冠脉综合征和 34例正常对照者中分离周围血单核细胞 ,抽提总RNA ,应用逆转录PCR差异显示检测。结果 PPARγ可在周围血单核细胞表达 ,且急性冠脉综合征患者周围血单核细胞表达显著下调 ;PPARγ表达水平与可溶性血管黏附分子 1、可溶性细胞间黏附分子和体重指数负相关 (r分别为 - 0 339、- 0 331和- 0 334 ,P <0 0 5 ) ,与载脂蛋白A1正相关。结论 急性冠脉综合征时单核细胞PPARγ表达下调 ,与血浆可溶性细胞黏附分子水平负相关 ,提示PPARγ可能参与急性冠脉综合征发病过程。 Objective To study the expression status of peroxisome proliferator activated receptors γ (PPARγ)in peripheral blood monocytes in acute coronary syndrome and its correlation with adhesion molecules. Methods Peripheral blood monocytes were isolated by Ficoll. Differential expression of PPARγ in peripheral blood monocytes was analyzed by RT-PCR. Results Expression of PPARγ in peripheral monocytes was significantly lower in acute coronary syndrome than in control (0.15±0.25 vs 0.83±0.31,P<0.01) . PPARγ was inversely associated with soluble vascular cell adhesion molecule-1 (r=-0.339, P<0.05), soluble intercellular adhesion molecule-1 (r=-0.331, P<0.05) and BMI (r=-0.334, P<0.05), and positively correlated with apoA1(r=0.289, P<0.05). Conclusions The expression of PPARγ is significantly reduced in peripheral monocytes in acute coronary syndrome,which is inversely associated with plasma soluble adhesion molecules. These results sugget that PPARγ may involve in the process of acute coronary syndrome.
