摘要
目的 研究建立血清孕酮固相试管酶免测定方法。方法 将抗孕酮抗体包被到固相试管壁 ,血清中的孕酮标记物竞争结合固相抗体 ,标本的浓度通过内插法从标准曲线上求得 ,并和磁酶免测定法平行对比测定。结果 该种方法和磁酶免测定法进行对比实验并作了直线回归分析 ,提示 2种方法高度相关 ,r=0 .932 ,P <0 .0 0 1;直线回归方程为Y =0 .5 79+0 .892X。两种疗法测定结果经配对t检验 ,P >0 .5 0 ,提示 2种检测方法无显著性差异。结论 该种孕酮固相试管酶免测定方法简便、准确 ,具有较高灵敏性和稳定性。
Objective To develop an assay for testing the concentration of progesterone in human serum by Enzyme immunoassay(polystyrene tube solid phase).Methods In which the assay we developed,a polyclonal antibody is conjugated to the polystyrene tube and alkaline phosphatase is used as a labelling enzyme.The progesterone in the sample reacted with the conjugated antibody and competes with a fixed amount of progesterone derivative conjugated to alkaline phosphatase.The concentration of progesterone can be determined directly by interpolation from the standard curve. Results The assay we developed was compared to encyme immunoassay(magnetic solid phase).A linear regression analysis waw performed indicating a high correlation.r=0.932,Y=0.579+0.892X.Paried samples test achieved:P=0.886,There was no significant difference between the two methods.Conclusions The method we established is rapid and accurate.It retained the same sensitivity and reliability of enzyme immunoassay(magnetic solid phase).
