甲基莲心碱对MCF-7细胞体外化疗增敏作用

Enhancement of cytotoxicity of anticancer drugs in vitro by neferine in MCF-7 cells

目的 :探讨甲基莲心碱 ( Neferine,Nef)的体外化疗增敏作用。方法 :以人乳腺癌细胞 MCF- 7为研究对象 ,MTT法测定药物的细胞生长抑制率 ,流式细胞仪 ( Flow cytometry,FCM)检测细胞凋亡。结果 :Nef( 5 um ol· L- 1 、10 um ol· L- 1 )分别与阿霉素 ( ADM)、5 -氟尿嘧啶 ( 5 - FU )、顺铂 ( CDDP)合用 ,可增加它们对 MCF- 7细胞的生长抑制率 ,q>1。Nef( 10 um ol· L- 1 可增强 10 umol· L- 1 ADM诱导 MCF- 7细胞凋亡的作用。结论 :Nef具有化疗增敏作用。

OBJECTIVE:To study the enhancemem of cytotoxicity of anticancer drugs in vitro by neferine(Nef).METHODS:Cytotoxicity assay was tested by MTT method,modulation of adriamycin(ADM)-induced apoptosis by Nef in MCF-7 cells was detected by Flow cytometty(FCM).RESULTS:The inhibitory rates(IR)of Nef (5umol·L -1 and 10umol·L -1 )combined with ADM,5-FU or CDDP were higher than those of these drugs used alone,and q values were more than 1.Nef(10umol·L -1 )promoted apoptosis induced by ADM in MCF-7 cells.CONCLUSlON:Nef increase sensitivity to anticancer drugs.
