摘要
目的 研究白细胞介素 6对人类免疫缺陷病毒 (HIV)gp1 2 0基因免疫小鼠后效果的调节作用。方法 构建表达中国流行株HIV 1gp1 2 0基因的核酸疫苗质粒pGP及共表达中国流行株HIV 1gp1 2 0基因与IL 6基因的核酸疫苗质粒pGPIL 6 ,并检测其在哺乳动物细胞中的表达。将上述两种质粒经胫前肌注射BALB/c鼠 ,检测gp1 2 0抗体的产生情况及ConA和LPS诱导的T细胞增殖能力。结果 以间接免疫荧光法 (IFA)检测到gp1 2 0在哺乳动物细胞中得到表达。pGPIL 6与pGP均可刺激小鼠产生抗gp1 2 0抗体 ,协同注射gp1 2 0和IL 6基因的小鼠产生的抗体滴度明显高于单独注射gp1 2 0的小鼠。pGPIL 6免疫鼠对ConA和LPS诱导的T细胞增殖能力明显高于单独注射gp1 2 0的小鼠。结论 协同应用IL 6基因可明显加强HIV 1gp1 2 0基因的免疫反应 ,为中国流行株HIV 1核酸疫苗的可行性提供了重要实验依据。
Objective To investigate the regulating effect of interleukin 6 (IL 6) gene on the immune response induced by human immunodeficiency virus(HIV)gp120 gene in mice. Methods The nucleic vaccine plasmid of HIV 1 CN gp120 gene and co expressive nucleic vaccine plasimd of pGPIL 6 were constructed, which containiag both HIV 1 gp120 and IL 6 gene. The two plasmids were then injected together into tibilias anterior muscles of BALB /c mice and the induced humoral immune responses were evaluated. Results IFA showed that the constructed plasmids were successfully expressed in mammalian cells. Mice immunized with both gp120 gene and IL 6 gene exhibited a significant increase in the production of anti HIV 1 CN gp120 antibody compared to the mice immunized only with pGP and the proliferating response of splenocytes to ConA and LPS elevated markedly. Conclusion Humoral immune responses of BALB/c mice to HIV 1 CN gp120 antigen can be enhanced when HIV 1 CN gp120 gene and IL 6 gene are used in coordination.
