华支睾吸虫成虫总RNA的一步法提取与纯化

TOTAL RNA ISOLATION FROM CLONORCHIS SINENSIS ADULT WORMS BY SINGLE-STEP METHOD

目的 为华支睾吸虫cDNA文库的构建提供基础。方法 采用硫氰酸胍 -酚 -氯仿一步法提取华支睾吸虫成虫总RNA。结果与结论 3 .5h左右可自华支睾吸虫成虫获得高纯度、完整性好的RNA。其总RNA存在体内缺口现象 ,2 8s电泳主带缺如。

Aim To provide the basis of the construction of C.sinensis cDNA library.Methods To extract and purify the total RNA from adult C.sinensis by guanidine thiocyanate-phenol-chloroform single step method.Results and Conclusion In about 3.5 hours,the intact C.sinensis RNA with high purity could be isolated.In agarose gel eletrophoresis,the total RNA of adult C.sinensis showed RNA in-vivo-nick phenomenon.It showed no 28s main band.
