丙烯腈对雄性大鼠生殖功能的影响 被引量:10

The effect of acrylonitrile on reproductive function of male rats

摘要 目的 探讨丙烯腈 (ACN)亚慢性染毒对雄性大鼠生殖功能的影响。方法 分别以 0、10、2 0、40mg/kgACN灌胃染毒雄性Wistar大鼠 13周 ,测定与精子形成有关的酶 ,并于染毒 12周末进行交配实验 ,分析ACN对雄性大鼠生殖功能的影响。结果 ACN染毒 13周 ,10mg/kg组乳酸脱氢酶(LDH)及其同工酶LDH X活力分别为 (2 .16 1± 0 .395 )U/mgpro和 (0 .2 89± 0 .16 3)U/mgpro ,比对照组[(1.82 2± 0 .2 6 3)U/mgpro和 (0 .185± 0 .0 5 4)U/mgpro]明显增高 ,差异有显著性 (P <0 .0 5 ) ,而酸性磷酸酶 (ACP)、碱性磷酸酶 (AKP)、山梨醇脱氢酶 (SDH)活力比对照组略有升高 ,但差异无显著性 (P >0 .0 5 ) ;40mg/kg组LDH及LDH X[(1.789± 0 .2 86 )U/mgpro和 (0 .15 0± 0 .0 35 )U/mgpro]、ACP、SDH活力与对照组比较 ,差异均无显著性 (P >0 .0 5 ) ;附睾中LDH及LDH X活力呈现与睾丸中一致的变化 ,但各组与对照组比较 ,差异亦无显著性 (P >0 .0 5 )。交配实验结果表明 ,各剂量组之间雌鼠受孕率、雄鼠生育指数、交配率差异均无显著性 (P >0 .0 5 ) ,剂量为 40mg/kg时 ,孕鼠每窝平均活胎数为 5 .88,明显低于对照组 (7.73) ,差异有显著性 (P <0 .0 5 ) ,胎吸收率为 19.6 6 % ,明显高于对照组 (7.6 1% ) ,差异有显著性 (P <0 . Objective To explore the effect of acrylonitrile(ACN) on the reproductive function of male rats. Methods Wistar male rats were daily administrated ACN by gavage 5 times a week for 13 weeks at the dose of 0,10,20,40 mg/kg body weight respectively.Mating experiment was conducted by the end of the 12th week.The activities of lactate dehydrogenase(LDH) and its isoenzymes LDH X,acid phosphatase(ACP),alkaline phosphatase(AKP),sorbitol dehydrogenase(SDH) in testes and epididymis in rats were detected. Results The 13 week treatment of ACN caused a significant increase in the activities of LDH and LDH X in testes[(2.161±0.395) U/mg pro and (0.289±0.163)U/mg pro] at the 10 mg/kg group as compared with control group [(1.822± 0.263 )U/mg pro and (0.185±0.054)U/mg pro],but the increase in activities of ACP,AKP,SDH showed no significant difference.With further increase of dose,a significant decrease in activities of LDH,LDH X,SDH and ACP was observed,but there was no difference in activities of such enzymes between 40 mg/kg group and control.In spite of the same changes in activities of LDH and LDH X in epididymis,there was statistical difference in activity of LDH only between 10 mg/kg group and 40 mg/kg group ( P <0.05).Mating experiment results indicated that ACN had no effect on sexual function of rats,and a decrease in numbers of living fetuses per litter (5.88) and an increase in rate of embryonic absorption (19.66%) at dose of 40 mg/kg as compared with control group( 7.73 , 7.61%) . Conclusion ACN could cause some alteration of enzyme in testes and epididymis,impair embryo by administrated male rats,so ACN may affect the male reproductive function.
