摘要
目的 建立适用于临床的N myc检测方法 ,提供预后信息。方法 应用荧光原位杂交 (FISH)技术检测N myc基因扩增 ,应用RT PCR和免疫组化技术分别检测N myc转录和蛋白水平表达。结果 N mycmRNA水平和N myc蛋白表达水平之间没有严格的平行关系。结论 FISH和免疫组化技术共同运用于N myc基因扩增和蛋白表达的检测 ,不仅能为NB预后和治疗提供更可靠的分子生物学信息 ,而且有助于进一步探讨N myc基因调节机制和N myc在NB临床行为中的作用。
Objective To evaluate the clinical significance of determination of N myc expression in Neuroboastoma and to establish the detection techniques. Methods FISH was used to observe N myc gene amplifiction, RT PCR and immunohistochemistry techniques were used to detect N myc mRNA and N myc protein respectively. Results FISH and immunohistochemistry techniques had the advantage of requiring a small amount of sample, less time consuming,being easy to perform and allowing analysis at the single cell level. They were suitable for clinical use. Conclusion The analysis of N myc gene amplification in combination with N myc protein expression not only provides molecular biologic information for clinical behaviors and treatment stratification but also contributes to further studies on mechanism of N myc gene regulation and the precise roles that N myc play in NB clinical behaviors
