摘要
目的 检测肝细胞癌组织和正常肝组织中端粒酶RNA成分 (humantelomeraseRNAcomponent,hTR)和蛋白催化亚单位 (humantelomerasereversetranscriptase ,hTERT)各自表达情况。方法 以反转录聚合酶链反应 (RT PCR)和套式聚合酶链反应 (nested PCR)分别检测肝细胞癌组织和正常肝组织中hTR和hTERT。结果 RT PCR反应不足以清晰检出hTR的设计扩增条带 ,在RT PCR基础上进行套式PCR ,则正常肝组织和肝细胞癌组织均检出hTR内引物扩增条带 ,只是在肝细胞癌组织中hTR表达量相对更高一些。hTERT仅在肝细胞癌组织中检出 ,正常肝组织中检测不到。结论 hTR在正常肝组织和肝细胞癌组织中均能检测到 ;hTERT与端粒酶活性间关系更密切 ,端粒酶的激活可能至少包括两个层次 :其一是端粒酶RNA成分的上调 ,其二是蛋白亚单位的表达。
Objective To examine the expression of hTR (human telomerase RNA component) and hTERT(human telomerase reverse transcriptase)in normal liver samples and hepatocellular carcinoma(HCC) samples.Methods hTR and hTERT were examined by RT PCR and nested PCR Results hTR was constitutively expressed both in HCC and normal liver samples, and hTERT mRNA was expressed only in HCC samples. Conclusion hTERT plays a critical role in determining the enzymatic activity of human telomerase, and up regulation of hTERT probably plays a role in the progression of human cancer.$$$$
