摘要
从湛江林科所赠送的感染的宿主幼虫中分离提取松茸毒蛾质型多角体病毒。多角体的形状为十二面体,大小差异较大,在0.6~1.1μ之间,病毒粒子为二十面体的球形颗粒,有12个突起,大小为65nm。经SDS-聚丙烯酰胺电泳,多角体蛋白由分子量27000的主带及18000的次带组成;病毒粒子蛋白由8条多肽组成,分子量在132000~49000之间。CPV蛋白富含天冬氨酸、谷氨酸及酪氨酸,而胱氨酸含量最少,具有典型的CPV指纹图谱,用SDS-热酚法抽提病毒核酸,表明为双链RNA,由10个基因片段组成,分子量在2.6×10~6~0.36×10~6道尔顿之间,总分子量为14.86×10~6道尔顿。
Dasychira axutha collenette cytoplasmic polyhedrosis virus (DaCPV) was isolated from Dasychira axutha Collenette in southern China and kindly provided by Zhanjiong Institute of Forestry.The purified DaCPV was studied by morphological and biochemical methods. The results showed that the polyhedrosis bodies were dodecahadron in morphology and quite variable in diameter from 0.6μ to 1.1μ. The vi-rions and icosahedron particles were 65 nm in diameter with 12 spikes. As determined by SDS-PAGE, the polyhedra protein contained 2 polypeptides 27,000 and 18,000 dalton in molecular weight respectively. The DaCPV virion had 8 polypsptides ranging from 132,000 to 49,000 dalton. Proteins from DaCPV were rich in aspartic acid, glutamic acid and tyrosine but poor in cystine and showed typical fingerprint.The viral nucleic acid was extracted by SDS-hot phenol method and determined as double stranded RNA composed of 10 genomic fragements ranging from 2.6×106 to 0.36×106 dalton in molecular weight and with a total molecular weight of 14.48×106 for the whole viral genome.
