RB、p53基因表达异常同大肠癌发生、发展的关系

The Significance of Over-expression in the RB and p53 gene on Colorectal Tumorigenesis

本文以大肠正常粘膜为对照，利用Ｎｏｒｔｈｅｒｎ印迹杂交、斑点杂交技术，检测了大肠癌、腺瘤中肿瘤抑制基因（ＲＢ和ｐ５３）的表达情况。实验结果表明：大多数大肠癌有ＲＢ基因、ｐ５３基因或二者同时存在的过表达现象，在个别腺瘤中也可见到这种现象。推测：可能由于ＲＢ、ｐ５３基因发生了点突变，导致基因产物失活，引起细胞增殖活动增强，反过来通过某种调节机制诱导ＲＢ基因出现过表达现象。

The expression of the RB and p53 gene in 17 colorectal carcinomas,10 adenomas and adjacent normal mucosas were observed,by employing Northern blot,Dot blot asay. Results showed that there is increase in the RNA level of either RB or p53 or both,in most carcinomas and some adenomas.It is predicted that a point mutation in the RB gene results in the production of an inactive protein and leads to increased cell proliferation,which,in turn,might induce RB gene over-expression through a regulatory mechanism.
