内皮素-1在暴发性肝功能衰竭发生中的作用

The action of endothelin-1 in fulminant hepatic failure in rats

目的 观察内皮素 (ET) - 1在暴发性肝功能衰竭发生中的作用。方法 以硫代乙酰胺 (TAA)灌胃染毒的方法建立大鼠暴发性肝功能衰竭模型 ,利用生化方法测定血浆内毒素及内皮素水平 ,采用 ET抗血清拮抗 ET- 1的作用。结果 TAA组血浆内皮素、内毒素和血清丙氨酸转氨酶水平显著高于正常对照组 (P<0 .0 5 ) ;TAA +ET- Ab组血浆内皮素、内毒素和丙氨酸转氨酶水平显著低于 TA A组 (P<0 .0 5 ) ,内皮素与内毒素水平呈显著正相关 (P<0 .0 1) ,内皮素抗血清可以拮抗 ET- 1的作用并可减轻肝坏死的程度。结论 内皮素可上调血浆内毒素水平 。

Objective To observe the action of endothelin 1 (ET 1) in fulminant hepatic failure (FHF). Methods The model of FHF in rats was set up by thioacetamide (TAA) per os,plasma endotoxin and endothelin were determined by using biochemical methods,anti ET 1 serum was adopted to check up the action of ET in FHF.Results The levels of plasma endotoxin,plasma endothelin and serum ALT activity in the TAA group were all higher than those of the normal group (P<0 05);the levels of plasma endotoxin,endothelin and serum ALT activity in the TAA+ET Ab group were all lower than those of the TAA group (P<0 05),the levels of plasma endotoxin and endothelin showed a positive correlation (P<0 01),hepatic necrosis was reduced by using anti ET 1 serum.Conclusion ET 1 induces the up regulating of the level of plasma endotoxin and correlates closely with the pathogenesis of FHF.
