用流式细胞计检测表皮生长因子对肝细胞DNA合成的刺激作用 被引量:5

摘要 用流式细胞术和单克隆抗溴脱氧脲嘧啶抗体标记 DNA 技术检测了表皮生长因子、胰高血糖素和胰岛素对大鼠初代培养肝细胞 DNA 合成的刺激作用。结果表明表皮生长因子对肝细胞DNA 合成具有明显的增强作用,并且这种作用可以被胰岛素进一步加强。当表皮生长因子、胰岛素和胰高血糖素三者联合使用时,肝细胞 DNA 合成显著增强。提示表皮生长因子,特别是它与胰岛素和胰高血糖素联合使用可以明显刺激肝细胞的再生。 The effects of epidermal growth factor (EGF),glucagon and insulin onthe DNA synthesis in primary culture of adult rat hepatocytes have been mea-sured using flow cytometry and monoclonal anti-BrdU antibody staining.Theresults showed that the DNA synthesis of the normal liver cells was in a stateof relative rest,the labelling index (L.I) was 0. 075% (4/5320) .When EGFwas added,the DNA synthesis rate increased (L.I.1. 43%).This stimulatoryeffect of EGF might be enhanced by insulin (L.I.163%).However,WhenEGF was administered in combination with glucagon and insulin,the DNAsynthesis was dramaticly increased (L.I.4. 18%,448/10730) .These suggestedthat EGF and EGF-glucagon-insulin therapy might be helpful in the treatme-nt of acute hepatic failure.As comparedto [~3H] thymidine technique,flow cytometry is more specificand can give a more correct result of DNA synthesis.It saves time and hasno radioactive hazard.
