摘要
成功地获得了既抗肾综合症出血热病毒,又抗辣根过氧化物酶的双特异性单克隆抗体(简称抗HFRSV-抗HRP BsMcAb)。方法是,先将分泌抗HFRSV McAb的杂交瘤A25-1细胞用杂氮鸟嘌呤进行诱变试验,建立了分泌抗HFRSV McAb的HGPRT细胞株(即HAT敏感型细胞株)251AGD6。用此细胞与HRP免疫的Balb/c小鼠脾细胞再次融合,得到分泌抗HFRSV-抗HRP的BsMcAb杂交杂交瘤(Hybrid Hybridoma)261-HRPB10细胞株。用双抗原免疫斑点试验和双抗原ELISA试验测定251-HRPB10腹水BsMcAb,滴度均为1000;用IFA测定单向抗HFRSV抗体滴度为10000;用免疫斑点法和ELISA测定单向抗HRP抗体滴度,均为100000。杂交杂交瘤的染色体数目在150条左右,证实是三细胞两次杂交瘤。
An hybrid hybridom, 251-HRPB10, which secretes anti-hemorrhagic fever with renal syndrome virus (HFRSV) and anti-horseradish peroxi- dase (HRP) bispecific monoclonal antibody (BsMcAb),was obtained.The titer determined for BsMcAb by bi-antigen dot immunobinding assay and bi-antigen sandwich. ELISA was 1 : 1 ,000. The titer determined for anti-HFRSV McAb by IFA was. 1:10,000. The titer determining for anti-HRP McAb by dot immunobinding assay and ELISA was 1:100,000. The chromosome number of the hybrid hybridoma 251-HRPB10 was about 150. Three peaks were found in 251-HRPB10 ascites fluids analysed by DEAE-cellulese. The first, second and third peaks were anti-HFRSV McAb anti-HFRSV-HRP BsMcAb and anti-HRP McAb respectively, the second peak being the highest. Production of BsMcAb against viruses was very important for the therapy and diagnosis of virus diseases.
