摘要
初步研究鸽视觉偏侧化的分子机制。建立鸽左侧单眼剥夺模型 ,采用抑制消减杂交和反Northern方法筛选右视盖差异表达的表达序列标签。自右视盖消减文库随机取 60个重组克隆进行反Northern杂交 ,证实为真阳性克隆 6个 ,随机对其一进行序列分析 ,发现该片段与人CDC10有 84 %的同源性。CDC 10属MBF转录因子复合体的一部分 ,介导细胞周期调节的转录 ,该表达序列标签在右视盖表达增强 ,可能调制一些视觉相关神经元的分化、增殖 。
Preliminarily to study the molecular mechanism of the visual lateralization in pigeons; the left monocular deprivation model of pigeons was established. By using suppression subtractive hybridization technique and reverse Northern assay , the differentially expressed sequenced tags (ESTs)for right optic tecta of pigeons were screened. 6 out of 60 ESTs were proved true positive by reverse Northern assay, 1 of them was sequenced , showing 84 % homology with homo sapien CDC10 in Genbank. CDC10 belongs to one part of transcriptionalfactor MBF complex, which mediats cell cycle regulated transcriptional activation at start. The high expression of this EST in right optic tecta suggests that it may modulate the proliferation and differentiation for some kinds of visual related neurons. The whole cDNA sequence of the EST should be cloned and its function should be investigated prior to elucidating the relationship between the pigeon visual lateralization and the gene.
