摘要
用酚氯仿去除重复缺氧鼠脑匀浆液中的蛋白质等大分子物质 ,以乳酸脱氢酶 (LDH)透出率为指标 ,观察去蛋白匀浆提取液对缺氧突触体的保护作用。重复缺氧 2 ,3次特别是 4次动物的去蛋白提取液显著降低缺氧突触体的LDH透出率 ;重复缺氧 4次组去蛋白匀浆提取液的LDH透出率亦显著低于纯氧对照组。结果提示通过缺氧预适应 ,鼠脑产生一种小分子活性物质 。
To observe protective effect of protein free brain homogenate taken from mice exposed repetitively to hypoxia on hypoxic insult of synaptosome, protein and macromolecular substances were removed by phenol chloroform from the homogenate, and efflux rate of lactate dehydrogenase(LDH) was measured as an index. The LDH efflux rate was found to be significantly decreased by the protein free homogenate of groups exposed to hypoxia for 2, 3, and 4 runs in particular. The LDH efflux rate in group exposed to hypoxia for 4 runs was also significantly lower than control group exposed to oxygen. The result indicates that an active substance with small molecule is produced in the brain of mice during hypoxic preconditioning and it posseses a role to protect the synaptome from hypoxia and self melt induced damage.
