摘要
目的 :探讨新生儿出生时淋巴细胞功能障碍的可能环节。方法 :给予新生儿脐血和成人外周血淋巴细胞抗CD3单克隆抗体、PMA和IM刺激后 ,应用3H TdR掺入法检测淋巴细胞的增殖反应 ;Westernblot法检测淋巴细胞PLCγ1的表达。结果 :抗CD3单克隆抗体刺激 ,脐血淋巴细胞增殖反应明显低于成人 ;PMA协同IM刺激后 ,脐血淋巴细胞的增殖反应明显高于应用抗CD3单抗刺激后的 ;脐血淋巴细胞PLCγ1的表达明显弱于成人。结论 :脐血淋巴细胞功能障碍可能与细胞活化的上游信号转导缺陷有关。
Objective:To explore the insufficiency mechanism of the cord blood lymphocytes of the neonates Methods: 3H Thymidine incorporation was used to analyze the proliferative response; Western blot technique was performed to assess PLCγ1 expression between cord and adult lymphocytes Results:The proliferative response of cord blood lymphocytes to anti CD3 mAb was significantly reduced compared to that of the adult's The proliferative response of cord blood lymphocytes to PMA+IM was significantly higher than the response to anti CD3 mAb The PLCγ1 expression was markedly lower than that of the adult's Conclusion:The lymphocytes dysfunction of the neonatal cord blood probably related to the signal transduction in the early stage lymphocytes activation
