摘要
目的 揭示海嘧啶的抗肿瘤作用机制。方法 采用流式细胞仪测定海嘧啶对人胃癌细胞凋亡及细胞周期的影响 ;采用激光扫描共聚焦技术观察海嘧啶对人胃癌细胞内 [Ca2 + ]i 的影响及 [Ca2 + ]i 变化时 Ca2 + 的来源。结果海嘧啶可诱导肿瘤细胞凋亡 ,可升高肿瘤细胞内 [Ca2 + ]i 的浓度 ,[Ca2 + ]i 升高时 Ca2 +来源于细胞外钙内流和细胞内钙释放。结论 海嘧啶的抗肿瘤机制为诱导肿瘤细胞凋亡 ,通过开放细胞膜钙通道和引起细胞内钙释放两条途径升高肿瘤细胞内 [Ca2 + ]i,启动细胞凋亡机制 。
Object To uncover the antitumor mechanism of sea pyrimidine (SP), a compound Chinese Western medicinal preparation containing 5 fluorouracil, extracts of Astragalus membranaceus (Fisch.) Bunge, Sophora flavescens Ait. and certain sea weeds. Methods Effect of SP on SGC 7901 gastric cancer cell cycle and apoptosis were determined by FCM and the source of Ca 2+ during intracellular [Ca 2+ ] i change, observed by laser scanning confocal microscopy. Results SP can induce apoptosis of SGC 7901 tumor cell, and elevate its [Ca 2+ ] i level. The source of Ca 2+ originated from extra cellular [Ca 2+ ] i and intracellular Ca 2+ release. Conclusion The antitumor mechanism of SP is its action to induce apoptosis of tumor cell through opening of calcium channel and release of intracellular calcium to initiate cellular degradation resulting in tumor cell apoptosis.
