紫杉醇诱导Lewis肺癌细胞凋亡的研究 被引量:1

摘要 目的 :探讨紫杉醇对Lewis肺癌细胞凋亡的影响。方法 :将Lewis肺癌细胞接种于C57BL/ 6小鼠皮下 ,将 2 0只小鼠随机分为对照组和紫杉醇组 ,分离皮下肿瘤结节 ,利用流式细胞计数仪检测细胞周期和凋亡率。采用免疫组化SP法利用图像分析仪检测bcl- 2蛋白 ,fas蛋白的表达。采用原位DNA末端标记法进行原位细胞凋亡的检测。结果 :(1 )对照组 (Ⅰ组 )凋亡率为 9 0 0 2 % ,G2 /M期细胞为 2 654。实验组 (Ⅱ组 )凋亡率为 2 5 772 % ,G2 /M期为 1 3 834 ,两者差异均有显著性 (P <0 0 1 )。 (2 )Ⅰ组细胞凋亡指数为 0 7% ,Ⅱ组细胞凋亡指数为 9 6 % ,两者差异有显著性 (P <0 0 1 )。 (3)Ⅰ组bcl- 2蛋白 ,fas蛋白的面积 /高倍视野分别为 1 73 52 0和 5 1 64 ,面密度分别为 1 1 7 42 8和 3 494,灰度分别为 2 2 1 39和 2 0 38。Ⅱ组bcl- 2蛋白 ,fas蛋白的面积 /高倍视野分别为 48 33和 55 780 ,面密度分别为 32 71 0和 37 750 ,灰度分别为 9 370和 2 3 1 4 0 ,Ⅰ组与Ⅱ组间bcl- 2蛋白 ,fas蛋白的差异均有显著性 (P <0 0 5)。结论 :紫杉醇可明显诱导Lewis肺癌细胞凋亡。 Objective:To investigate the effects of taxol on celluar apoptosis of Lewis lung cancer (LLC).Methods:LLC cells(1 25×10 6/mouse)were inoculated subcutaneously to 20 mice,which were randomized into control group (group 1) and taxol group (group 2) and given respective treatment consecutively in 5 days after inoculation.For group 1 natrii chloride was injected for 10 days,for group 2 taxol was injected for 10 days.On the 18th day,all the mice were killed.In siut endlabeling (ISEL) was adopted to inspect apoptosis.Immunohistochemist SP was adopted to inspect expression of bcl-2,fas protein by image analysis.Results:(1)Apoptosis rate of group 1 is 9 002,G 2/M stage cell is 2 654;Apoptosis rate of group 2 os 25 772,G 2/M stage cell is 13 834,significant difference was observed between the two groups (P<0 01).(2)Apoptosis index of group 1 is 0 7%;Apoptosis index of group 2 was 9 6%,significant difference was observed between the two groups (P<0 01).(3)In group 1,the area/HP of bcl-2 protein,fas protein were 173 520,5 164 respectively;The area/HP of bcl-2 protein,fas protein were 173 520,5 164 respectively;The area density of bcl-2 protein,fas protein were 117 428,3 494 respectively;The gray scale resolution of bcl-2 protein,fas protein were 22 139,2 038 respcetively.In group 2,the area/HP of bcl-2 protein,fas protein were 48 330,55 780 respectively;The area density of bcl-2 protein,fas protein were 32 710,35 750 respectively;The gray scale resolution of bcl-2 protein,fas protein were 9 370,23 140 respectively.bcl-2 protein and fas protein in the two groups had significant difference (P<0 05).Conclusions:Taxol can remarkably induce apoptosis of LLC which provides theoretical basis for clinical treatment of human lung cancer
