摘要
目的 用重症肌无力 (MG)患者的乙酰胆碱受体抗体 (ACh Rab)选择性地显示神经元烟碱型乙酰胆碱受体 .方法 用 Elisa法测定 MG患者血中的 ACh Rab滴度 ,然后用硫酸铵盐析和免疫亲和层析法提取纯化 ACh Rab,再用免疫组化法探讨 ACh Rab与大鼠 CNS神经元 - n ACh R之间的免疫结合反应 .结果 首次表明 ACh Rab与神经 - n ACh R之间的阳性免疫结合反应广泛分布于鼠 CNS许多部位 ,如大脑皮层、脑干各颅神经运动核团、脊髓运动神经元等部位 ,与用其他方法显示者其分布一致或类同 .结论 MG患者 ACh Rab可与大鼠和猴 CNS神经 - n ACh R交叉结合 ;可用于选择性地显示 CNS神经 - n ACh R.
AIM To study the method of expression of neuronal nicotinic acetylcholine receptor by acetylcholine receptor antibody purified from myasthenia gravis serum. METHODS ELISA assay was applied to detect the acetylcholine receptor antibody (AChRab). AChRab was purified by immunoaffinity tomography method and was used to study the immunoreaction between acetylcholine receptor antibody and rat central nervous system neuronal nicotinic acetylcholine receptor.RESULTS The positive immunoreaction between acetylcholine receptor antibody and central neuronal nicotinic acetylcholine receptor was widely distributed in rat central nervous system, for instance, cerebral cortex, brain stem motor necleus and spinal motor neurons. The distribution pattern was consistent with other methods. CONCLUSION AChRab from MG can crossreact with rat neuronal nicotinic acetylcholine receptor and can selectively express central neuronal nicotinic acetylcholine.
