bFGF保护脊髓组织、促进神经再生的实验研究 被引量:1

摘要 目的 探讨应用外源性碱性成纤维细胞生长因子 (basicfibroblastgrowthfactor,bFGF)对脊髓损伤的保护作用以及促进脊髓神经再生的影响。方法 利用Allen氏WD (Weightdrop ,WD)技术 ,以 10 g× 2 5cm致伤力造成SD大白鼠T8脊髓损伤模型 ,并于损伤平面以下蛛网膜下腔置细塑料导管。治疗组分别于术后即刻、 1、 2、 3、 4、 8、 12、 2 4及 48h经导管注入bFGF溶液 2 0 μl (含bFGF10 0 μ) ,以后每周经导管注入 2 0 μlbFGF ;对照组则在同时间注入等量生理盐水 ,术后取伤区脊髓组织进行形态学观察及特殊染色 ,观察神经纤维再生情况。结果 伤区白质内髓鞘结构紊乱 ,囊性变严重 ,应用bFGF可明显改善 ;Gless染色发现bFGF治疗组神经纤维生长情况明显优于生理盐水对照组 ,Fast-blue染色发现bFGF治疗组脊髓变性较轻 ,经统计学分析两组差异有显著性意义 (P <0 0 1)。结论 脊髓损伤后应用外源性bFGF可以保护脊髓神经组织 ,并且对神经纤维有明显的促进作用。 Objective To study the protecive effects and nerve regeneration of basic fibroblast growth factor on spinal cord injury.Methods Twenty-four SD rats with 10 g×2.5 cm impact to T 8 SCI were divided into two groups,and a thin plastic tube was situated in subarachnoid space below the injury level for perfusion.bFGF-treated animals received bFGF 100 μ(20 μl liquid)from the tube at once、0.5、1、2、3、4、5、6、12、24 and 48 hours after injury,and these rats received bFGF 20 μl from the tube once a week.The saline treated animals received the equal volume of normal saline at the same time.The injured spinal cord was detected by morphological observation and the regeneration of axons in injured spinal cord was observed by Glees and Fastblue staining in eight weeks later after injury.Results The damages of gray and white matter in bFGF-treated rats were slighter than those of saline-treated rats,the numbers of axons in the injured spinal cord reduced and microcysts increased in the saline-treated rats,whereas in bFGF-treated group the numbers of axons increased significantly.Conclusion bFGF plays an important role in nerve regeneration of spinal cord after spinal cord injury.
