摘要
本文主要研究低分子量的硫醇类化合物———β 巯基乙醇 (β ME)对水牛胚胎体外发育的影响。通过在水牛的体外成熟、受精后胚胎的体外发育过程中分别添加 5 0和 10 0 μM的 β ME ,来检测 β ME的添加对水牛受精卵的分裂率和植入前胚胎体外发育的影响。结果表明 ,对照组 (0 μM )与实验组 (5 0和 10 0 μM)相比 ,4 8h受精分裂率 (分别为 4 5 .88%、4 6.38%和 5 0 .67% )差异不显著(P >0 .0 5 ) ,但囊胚率 (分别为 8.97%、2 6.61%和 2 6.5 5 % )差异极显著 (P <0 .0 0 1) ,而孵化囊胚率(分别为 80 .0 %、90 .4 8%和 83.33% )差异不显著 (P >0 .0 5 ) ,在两个实验组 (5 0和 10 0 μM )之间分裂率、囊胚率以及孵化囊胚率均为差异不显著 (P >0 .0 5 )。结果表明在体外培养条件下添加一定浓度的 β ME可以显著促进更多的分裂后水牛早期胚胎发育至囊胚阶段 。
This study investigated the effects of β Mercaptoethanol (β ME), a low molecular weight thiol compound, on the in vitro development of water buffalo embryos. β ME(50μM and 100μM,treatments) was added in the media during in vitro maturation and in vitro culture but not in vitro fertilization of water buffalo oocytes or embryos. Compared with the group without β ME (0μM, control),this was no difference (P<0.05) in cleavage rates at 48 hrs (45.88% Vs 46.38% and 50.67%) and percentage of hatched blastocysts (80.0% Vs 90.48% and 83.33% ), however, a highly significant difference (P<0.01) was obained in percentage of blastocysts (8.97% Vs 26.61% and 26.55%) between control group (0μM) and treatment groups (50μM and 100μM). Insignificant differences (P>0.05) were observed in cleavage rates, percentage of blastocysts and percentage of hatched blastocysts between two treatment groups. These results indicate that the addition of β ME in IVM and IVC media improves remarkably the development of water buffalo embryos derived from IVM-IVF to the blastocyst stage.
