摘要 目的 :研究小鼠胚胎质量对其分泌 EGF、TGF-β1、IGF- 的影响。方法 :取小鼠 2 -细胞胚胎 ,分为正常和异常两组 ,体外培养至囊胚期 ,对胚胎的形态学指标进行观测 ;检测胚胎培养液中 3种生长因子的含量并进行比较。结果 :正常组胚胎培养液中 EGF、IGF- 的含量显著高于异常组 ,且 EGF含量与胚胎生长发育的形态学指标呈正相关 ,但与着床指标相关不显著 ;IGF- 与胚胎生长发育及着床指标均呈正相关。TGF-β1的含量两组间差异无显著性。结论 :胚胎培养液中 EGF、IGF- 含量测定可反映胚胎生长发育能力 ,IGF- 还可反映胚胎着床能力 ,从而有利于了解胚胎功能状态及生存潜力 ,为胚胎质量的检测提供了一条新的思路。 Objective:To study the effect of mouse embryo quality on the secreting levels of epidermal growth factor(EGF), transformine growth factor-β 1 (TGF-β 1) and insulin-like growth factor-Ⅰ(IGF-I). Methods: Two cell mouse embryos, normal and abnormal, were cultured to blastocyst stage. Their morphologic indices were determined. The level of EGF, TGF-β 1, and IGF-II in the culture media were measured. Results: 1. The levels of EGF and IGF-II of the normal group were significantly higher than those of the abnormal. The level of EGF was positively related to morphologic indices of embryo development, but was not related to the implantation indices. The level of IGF-II was positively related to indices of both embryo development and implantation. 2. The difference in the levels of TGF-β 1 between two groups was not statistically significant. Conclusion: The levels of EGF and IGF-II in embryo culture media may reflect the capability of embryo development, IGF-II may also reflect embryo embedding capability. It might be useful in assessment of embryo functional status and its vital potential.
