摘要
建立了一种在Nova -PakCN HP氰基柱上拆分 1 α 萘氧基 3 异丙氨基 2 丙醇对映体的高效HPLC方法。流动相组成为V(二氯甲烷 )∶V(异丙醇 )∶V(乙酸 ) =98∶1∶1,添加N 苄氧羰基 S 苯基 L 半胱胺酸 (BPC)为手性离子对试剂 ,c(BPC) =3.0mmol/L ,流动相流速为 0 .3mL/min ,检测波长为 2 5 4nm ,1 α 萘氧基 3 异丙氨基 2 丙醇对映体的分离选择性为 1.15 ,分离度为 1.19,线性范围为 0 .1~ 4.5mg/mL ,浓度测定的变异系数为 0 .33 %~ 0 .71%
The resolution of enantiomers of 1-α-naphthyloxy-3-isopropylamino-2-propanol was studied by means of HPLC. Dichloromethane-isopropanol-acetic acid was used as mobile phase[V(dichloromethane):V(isopropanol):V(acetic acid) = 98:1:1], containing N-benzyloxycarbonyl-S-phenyl-L-cysteine (BPC) as chiral ion pair agent[c(BPC) = 3.0 mmol/L], at flow rate of 0.3 mL/min in Nova-Pak CN-HP column with UV-detector of 254 nm. This method gave separation selectivity of 1.15, separation degree of 1.19, linear ranger of 0.1-4.5 mg/mL and coefficient of variation of 0.33% - 0.71%.
