摘要
目的 探讨碱性成纤维细胞生长因子 (bFGF)和转化生长因子 β1(TGFβ1)在人体超负荷性心肌肥厚发生机制中的作用。方法 用天狼猩红、免疫组织化学及原位杂交技术检测了 32例肥大心肌 (左、右心室心肌各 16例 )组织中胶原、bFGF和TGFβ1mRNA和蛋白的表达情况。 结果 压力和容量超负荷都可使心肌组织胶原增生 ,bFGF、TGFβ1mRNA和蛋白表达水平显著增加。bFGF、TGFβ1mRNA和蛋白表达水平与心肌细胞横径增加呈显著正相关 ,TGFβ1mRNA和蛋白表达水平与心肌胶原总量也呈显著正相关。结论 bFGF和TGFβ1表达可能是介导人体超负荷性心肌肥厚发生的因素之一。
Objective To investigate the role of basic fibroblast growth factor (bFGF) and transforming growth factorβ1 (TGFβ1) in the process of human heart hypertrophy induced by overload. Methods Sirius Red staining, immunochemistry and in situ hybridization were used to detect collagen, bFGF and TGFβ1 gene expression in 32 human heart hypertrophy(right and left ventricles,each 16 cases). Results The total collagen in hypertrophic myocardium was 1.5 times the number of normal heart ( P < 0.05 0.01), and the rate of collagen I and III was also increased 2.1 times as compared with control. bFGF and TGFβ1 mRNA expression were elevated significantly in hypertrophic myocardium (bFGF,left /right ventricle: 0.28±0.04; 0.18±0.03/ 0.29± 0.05; 0.20±0.03, P <0.01; TGFβ1,left /right ventricle: 0.26±0.05; 0.20±0.02/ 0.25±0.06;0.18±0.02, P <0.05),bFGF and TGF β1 protein expression were also increased markedly (bFGF,left /right ventricle: 0.27±0.03; 0.19±0.05/ 0.28±0.03; 0.18±0.03, P <0.01; TGFβ1,left /right ventricle: 0.22±0.03; 0.16±0.02/ 0.23±0.03; 0.17±0.04, P <0.01). bFGF and TGFβ1 mRNA and protein expression level were positively related with left and right myocardial hypertrophy, and TGFβ1 mRNA and protein expression were also positively with total collagen of hypertrophic myocardium. Conclusion It is suggested that bFGF and TGFβ1 mRNA and protein expression may play a role in the mechanism of human heart hypertrophy induced by overload.
