猕猴桃基因组DNA的提取及其RAPD扩增研究 被引量:32

摘要 采用 5种不同的方法提取猕猴桃基因组DNA ,并将 5种方法所得DNA用于RAPD扩增。结果表明 ,改进的CTAB法和氯化苄法提取DNA的得率最高 ,分别达 30 .6 μg/mg干样和 32 .7μg/mg干样 ,是经典CTAB法和高盐低pH法得率的 3倍 ,SDS法得率的 9倍。改进的CTAB法和氯化苄法提取的DNA扩增效果最好。 The genomic DNAs from ll breeds of Actinidia chinensis planch were extracted by 5 DNA extraction methods,and those DNA templats were used in RAPD reaction,respectively.The results suggested that the yields of DNA extracted by improved CTAB method and benzyl chloride method were three times that of conventional CTAB method,and method with low pH extraction mediun and high salts method,and 9 times that of SDS method.The DNA templats extracted from Actinidia chinensis planch.by improved CTAB method and benzyl chloride method were quite good for RAPD reaction.RNA,no matter what deprived or not from the DNA templates,had no influence on amplification.
