5种白蚁基因组DNA随机扩增多态性研究 被引量:6

摘要 采用 RAPD(Random amplified polymorphic DNA)技术对尖唇异白蚁 (H eterotermesaculabialis)、黄肢散白蚁 (Reticulitermes flaviceps)、扩头散白蚁 (R.ampliceps)、细颚木鼻白蚁(Stylotermes angustignathus)和陇南树白蚁 (Glyptotermes longnanensis)等 5种白蚁的基因组DNA多态性进行了初步分析。在所用的 2 0个随机引物中有 4个引物能扩增出稳定的带型 ,5种白蚁 6个群体共扩增出 43条带 ,均为多态性片段 ;同一种类不同群体和同一群体不同品级间存在一定程度的多态性 ,但种间扩增片段差异显著大于种内差异。以 UPGMA方法将扩增片段分布和共享度数据矩阵进行聚类分析 。 Genetic variation in five species of termites, Reticulitermes flaviceps, R. ampliceps, Stylotermes angustignathus and Glyptotermes longnanensis was detected using randomly amplified polymorphic DNA (RAPD) markers. Of the 20 arbitary 10 base primers tested, 4 (20%) were informative and yielded 43 bands that were all polymorphic. Polymorphism was found in different populations of the same species and in different castes of one colony, but the difference of the amplified fragments in interspecies was significantly bigger than that of the intraspecies. The relationship indicated by the cluster analysis (unweighted pair group method with arithmetic mean, UPGMA) suggested that Heterotermes aculabialis should be transferred to Reticulitermes as originally combined.
