表阿霉素和阿霉素诱导人肝癌细胞凋亡 被引量:12

摘要 目的 观察和比较表阿霉素和阿霉素诱导肝癌细胞凋亡的作用 ,为临床治疗肝癌的药物选择提供依据 .方法 用 2 0 μmol· L- 1表阿霉素和阿霉素分别作用于人肝癌细胞株HCC- 92 0 4细胞 3h,换液继续培养 8h.用细胞形态学观察、DNA电泳和流式细胞仪判定细胞凋亡情况 .结果 表阿霉素和阿霉素分别作用于 HCC- 92 0 4细胞 11h后 ,细胞形态学改变均符合凋亡细胞特征 .凋亡细胞分别占细胞总数的13.8%和 13.1% .MTT试验示各实验组与空白对照组间比较相差显著 ,表阿霉素和阿霉素相同剂量处理组之间比较无显著差异 .细胞周期检测表阿霉素组 G1 期、G2 期和 S期分别为 5 6 .4% ,10 .4%和 38.2 % ,阿霉素组分别为 6 1.1% ,8.6 %和 30 .3% ,对照组为 31.1% ,6 1.9%和 6 .0 % .结论 表阿霉素和阿霉素均能有效地诱导人肝癌细胞株 HCC- 92 0 4细胞发生凋亡 ,无明显差异 . AIM To provide the possibility for choice of drugs in clinical therapy of hepatic cancer, we observed and compared the effects of epirubicin and doxorubicin on inducing apoptosis in hepatic cancer cells. METHODS Human hepatic cancer cell strain were cultured with epirubicin (20 μmol·L -1 ) and doxorubicin (20 μmol·L -1 ) for 3 h respectively. Then the medium was replaced with a fresh one, and the cells were continuously cultured for 8 h. The apoptosis were identified and analyzed by cyto morphology, DNA electrophoresis and flowcytometry. RESULTS MTT assay indicated a significant difference between the experimental groups and the blank control group. The percentage of apoptotic cell was 13.8% in epirubincin group, and 13.1% in doxorubicin. No significant difference was observed between the groups treated with equal concentrations of the two drugs. Morphological changes in HCC 9204 cell, which was observed after 11h continuous culture with the two drugs indicated following apoptotic traits: ① DNA electrophoresis showed ladder patterned bands; ② Flowcytometry identified peaks typical of apoptosis. Cells in G 1, G 2 and S phase of cell cycle were 56.4% , 10.4% and 33.2% in epirubincin group, and 61.1% , 8.6% and 30.3% in doxorubicin group, and 31.1% , 61.9% , 6.0% in control group, respectively. CONCLUSION The apoptosis can be induced effectively by both epirubicin and doxorubicin in HCC 9204 cell, that shows little significant difference in DNA electrophoresis, flowcytometry and the morphological obscervation.
