兔肾生长刺激物质治疗急性肾功能衰竭

摘要 目的 从大白兔胎肾细胞内提取、部分纯化治疗急性肾功能衰竭 (ARF)的有效物质 ,为胎肾细胞移植提供实验依据 .方法 制备胎肾内生长刺激物质 ,经过盐析、透析、液相色谱分离提取、部分纯化 ,进行动物实验 ,3H- Td R参入测定其 Bq.结果 胎肾细胞移植治疗 ARF的有效成分是 Mr2 80 0 0耐热的蛋白质 . 70 0 g· L- 1 以上的硫酸胺能沉淀其活性成分 ,离子交换色谱 , 峰为活性峰 ,凝胶过滤色谱 , 峰为活性峰 .结论 胎肾细胞移植治疗 ARF的有效成分是 Mr2 80 0 0耐热的具有活性的细胞因子 。 AIM To extract and purify effective substance in foetus renal cell from large white rabbit, which could be effective to acute renal failure (ARF). METHODS Growth stimulant on foetus renal cell was drawn and purified by salting out method, dialysis and liquid chromatography (LC). Activity was determined by animal experiment. Bq value was determined by 3H TdR incorporation. RESULTS The available composition which could be effective to ARF in foetus renal cell was an thermostabile protein whose molecular weight was M r 28 000 . The active mass were precipitated in 700 g·L -1 ammonium sulfate. The active peaks were peak Ⅳ, Ⅴ in ion exchange chromatography and peak Ⅰ, Ⅱ in gel filtration chromatography. CONCLUSION The available composition which could be effective to ARF in foetus renal cell is an active thermostabile cellular factor which could be only or mostly existent in foetus renal cell.
